Assay Principles

 

The micronucleus assay detects aberrant, small nuclei present in the cell cytoplasm. Micronuclei may contain whole chromosomes ‘lost’ during mitosis or fragments generated by chromosome breakage. Usage of the micronucleus assay has increased in recent years – it is faster and less technical than the analysis of chromosomal aberrations. However, the traditional manual scoring method is laborious and so Litron Laboratories’ flow cytometric method is a significant advance. Flow cytometric assessment permits automated scoring, which removes subjectivity, and rapid measurement of large numbers of cells increases the statistical robustness of the results. In addition, the Litron method allows simultaneous collection of relative survival and membrane integrity parameters for cytotoxicity assessment, as well as cell cycle information.  The service from Gentronix allows testing both in the presence and absence of exogenous metabolic activation using S9 fraction.
