96 Well Plate Assays:
Raw absorbance and fluorescence data taken from the microplate reader is swiftly and automatically processed by the Excel based GreenScreen software provided with each kit. An example of the results output for a single microplate run using the GreenScreen assay is shown below and is similar for GreenScreen GC and HC, both based on a 96 well microplate format. Four test compounds can be run in each 96 well microplate together with appropriate genotoxic and cytotoxic standard compounds and untreated controls. Each test chemical is usually run over nine twofold serial dilutions and the genotoxicity and cytotoxicity results are automatically processed, interpreted and displayed as shown below. The multi-layer output provides (i) a single sheet summary of the genotoxicity and cytotoxicity results for all 4 compounds, plus the standard compound results for intra-plate quality control, (ii) detailed graphical results for each compound for user inspection, (iii) raw and processed numerical data for added transparency of the results.
Cuvette Based Assays:
Raw absorbance and fluorescence data are recorded from the portable cuvette based instrumentation and entered into to the software template, where they are automatically processed. The software gives a semi-quantitative assessment of genotoxicity and cytotoxicity for each of 6 cuvette samples per run. Where the run comprises 6 dilutions of the same sample a graphical output of any dose dependent trends in genotoxicity and cytotoxicity are displayed and quantitative genotoxicity (LOEC) and acute toxicity (EC50 and LOEC) results are calculated. Results for standard genotoxic and cytotoxic compounds and untreated controls are also presented.
